SUMMARY

Aims: This study aimed at establishment of CTOS (cancer tissue-originated spheroid) panel covering intra-patient heterogeneity for colorectal cancer, lung cancer and small cell carcinoma of the cervix. With the panel, changes in preparation, culture, and xenograft were pathologically, biochemically, and molecular biologically monitored.

Methods: Exome analysis by using Illumina HiSeq 2000

Participants/Materials: Colorectal cancer, Lung cancer, Small cell carcinoma of uterine cervix

colorectal cancer: 8 cases, lung cancer: 2 cases, small cell carcinoma of uterine cervix: 2case

 1. blood cells: 12 samples

(colorectal cancer: 8 samples, lung cancer: 2 samples, small cell carcinoma of uterine cervix: 2samples)

 2. CTOS (primary culture): CTOS derived from patient tumors: 5 samples

(colorectal cancer: 2 samples, lung cancer: 2 samples, small cell carcinoma of uterine cervix: 1 sample)

 3. CTOS (subculture):

  a. CTOS prepared from a CTOS (the same as 2)-derived mouse xenograft: 14 samples

   1st subculture: 1 sample (colorectal cancer: 1 sample)

   2nd subculture: 4 samples (small cell carcinoma of uterine cervix: 1 sample, colorectal cancer: 3 samples)

   3rd subculture: 5 samples (colorectal cancer: 3 samples, lung cancer: 2 samples)

   5th subculture: 4 samples (colorectal cancer: 2 samples, lung cancer: 2 samples)

  b. CTOS (the same as 2) cultured in vitro: 3 samples (lung cancer: 2 samples, colorectal cancer: 1 sample)

   2nd subculture: 1 sample (lung cancer: 1 sample)

   5th subculture: 2 samples (lung cancer: 1 sample, colorectal cancer: 1 sample)
