SUMMARY

Aims: Human whole saliva (WS) contains two types of exosomes (exosome I and exosome II) that are small membrane vesicle (30-100 nm in diameter). To clarify the host defense functions of exosomes, we investigated long RNA components (protein-coding RNAs and long non-coding RNAs) of exosome I, exosome II, and WS by next generation sequencing.

Methods: Human whole saliva was collected by discharge. Long RNA components (protein-coding RNA and long non-coding RNA) of exosome I, exosome II, and WS were analyzed by next generation sequencing.

Participants/Materials: Human whole saliva was collected from one Japanese healthy female volunteer with written informed consent.

Cell Lines -
Library Construction (kit name) Isolated RNA samples from exosome I, exosome II, and WS were amplified using the RampUP RNA Amplification Kit (Genisphere, PA, USA). cDNAs were generated using the amplified sense RNA, according to the manufacturer (Illumina Inc., CA, USA). The cDNA fraction in the range of 200–400 nucleotides was extracted and amplified by PCR.
